海带对实验性高脂血症大鼠降血脂作用及其机制 被引量:9

EFFECTS OF LAMINARIA JAPONICA ON SERUM LIPID IN RATS WITH EXPERIMENTAL HYPERLIPEMIA

摘要 目的探讨海带对实验性高脂血症大鼠血脂的调节作用及其机制。方法健康雌性Wistar大鼠40只,随机取10只作为正常对照组,普通饲料喂养;余30只应用高脂饲料喂养建立高脂血症动物模型。喂养4周后将造模成功的24只大鼠随机分为模型对照组、辛伐他汀组和海带组,各8只。模型对照组普通饲料继续喂养2周;辛伐他汀组普通饲料喂养,给予辛伐他汀10mg/kg(生理盐水稀释至1mL)灌胃,每日1次,共2周;海带组给海带粉饲料喂养2周。检测各组大鼠血清三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)、丙二醛(MDA)、一氧化氮(NO)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、脂蛋白酯酶(LPL)和肝酯酶(HL)的活性。结果辛伐他汀组和海带组动物血清TG、TC和LDL水平较模型对照组均显著下降,而HDL显著升高(F=4.22~37.04,q=4.76~13.57,P<0.05);海带组与辛伐他汀组血清TG、TC和LDL水平比较差异无显著性(q=0.31~3.15,P>0.05)。辛伐他汀组和海带组动物血清LPL和HL水平均显著高于正常对照组和模型对照组(F=68.79、92.12,q=5.51~19.09,P<0.05)。辛伐他汀组和海带组动物血清MDA和NO水平较模型对照组均显著下降(F=8.24、39.01,q=4.66~14.32,P<0.05),而血清SOD和GSH-PX活性均显著高于模型对照组(F=21.23、31.61,q=9.20~11.21,P<0.05)。海带组与辛伐他汀组比较,MDA、NO、SOD和GSH-PX等指标差异均无显著性(q=0.95~2.69,P>0.05)。结论海带可能具有类似辛伐他汀类药物的作用,通过增强LPL和HL的活性,影响TG、TC、LDL和HDL等血脂组分的代谢,而发挥调节血脂水平的作用。 Objective To investigate the effects of laminaria japonica(L.japonica)on serum lipid of experimental hyperlipemia in rats.Methods Forty healthy female Wistar rats were involved in this study,and 10 were selected from among them in random assigned as normal controls to be fed with ordinary forage,and the rest 30 were offered high-fat diet to create a hyperlipemia(HL)model in rats.After four weeks of feeding,24 rats with HL were equally randomized to model-control group,Sim-vastatin group and L.japonica group.For model-control group,ordinary forage was continued;for Simvastatin group,the drug was given intragastrically,10 mg/kg diluted in 1ml normal saline,once a day;for those in L.japonica group,the diet mixed with L.japonica powder was supplied,the course was two weeks for all the three groups.The serum triglyceride(TG),total cholesterol(TC),low-density lipoprotein(LDL),high-density lipoprotein(HDL),malondialdehyde(MDA),nitric oxide(NO)and the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX),lipoprotein lipase(LPL)and hepatic lipase(HL)were measured.Results The serum TG,TC and LDL in both Simvastatin and L.japonica groups were lower than that in the control,while the serum HDL elevated(F=4.22-37.04,q=4.76-13.57,P〈0.05),but no significant differences of that were found between Simvastatin group and L.japonica group(q=0.31-3.15,P〈0.05).The serum LPL and HL of rats in Simvastatin group and L.japonica group were markedly higher than those in the normal control and model-control group(F=68.79,92.12;q=5.51-19.09;P〉0.05).The serum MDA and NO in Simvastatin group and L.japonica group were significantly lower than that of the model-control(F=8.24,39.01;q=4.66-14.32;P〈0.05),while the activity of serum SOD and GSH-PX was significantly higher(F=21.23,31.61;q=9.20-11.21;P〈0.05).A comparison between Simvastatin and L.japonica groups showed no significant differences in MDA,NO,SOD and GSH-PX were noticed(q=0.95-2.69,P〉0.05).Conclusion L.japonica may have an effect similar to Simvastatins,which affects the metabolism of TG,TC,LDL and HDL to educe a regulation of blood lipid by enhancing the activity of LPL and HL.
