摘要
目的研究闪光视觉诱发电位(F-VEP)在监测实验性自身免疫性脑脊髓炎(EAE)视神经病变中的作用。方法制作Wistar大鼠EAE模型,以对照组大鼠F-VEP检测值确定正常参考值范围。模型组大鼠于豚鼠脊髓匀浆乳化剂免疫后不同时间点进行F-VEP检测,发病大鼠于发病后第6天行电生理检查后取视神经、脑、脊髓等通过伊红(HE)染色和LFB-焦油紫染色法进行病理检查。结果发病大鼠均出现不同程度F-VEP异常和视神经病理表现异常:F-VEP异常主要表现为P波和/或N1波潜伏期延长,最长延长分别为59.58%和35.33%;视神经病理改变主要表现为髓鞘脱失和炎性反应。F-VEP检测大鼠视神经病变的灵敏度达到100%,特异度为50%。结论 F-VEP可用于评价视觉系统损伤。
Objective To explore the effect of flash visual evoked potentials (F-VEP) in monitoring optic neuropathy in a rat model of EAE. Methods Established a Wistar rat model of EAE with guinea pig spinal cord homogenate and recorded F-VEP in the control group as normal reference value for this study. The rats in experimental group were performed F-VEP tests at different time points after immuno-inoeulation. Animals were executed when the electrophysiologieal tests were performed, brains and spinal cords were removed for pathological analysis (with HE and LFB staining methods). Results Disease incidence of EAE was 90% in experimental group. Distinct degrees of dysfunctions in electrophysiological responses as well as histological evidences in optic nerves were observed. F-VEP changes were characterized by prolonged latencies of P waves and N1 waves. Latencies of P and N1 waves were maximally prolonged by 59.58% and 35.33o/00, respectively. Optic neuropathy mainly manifested as demyelination and inflammation. Sensitivity of F-VEP detecting optic neuropathy in EAE rats reached 100%. Conclusions F-VEP can be an operative indication during evaluating optic nerve involvement and monitoring pathological course of EAE.
