摘要
目的:探讨同一温度不同时间和同一时间不同温度加热对凋亡坏死、survivin、caspase基因表达的影响及其作用机制。方法:对数生长期肺癌A549细胞,分别在同一温度不同时间和同一时间不同温度加热,用流式细胞仪检测细胞凋亡百分比和坏死百分比,用RT-PCR检测survivin和caspase基因的表达。结果:加热后细胞凋亡增加,加热温度42℃和加热时间2h最为明显。随加热温度升高和加热时间增加坏死增加。加热明显加强Casepase-3的表达和降低Survivin的表达,且42℃加热不同时间时,2h效果最明显,不同温度加热1h时42℃效果最明显。结论:(1)加热能明显抑制肺癌细胞生长,促进肺癌细胞的凋亡。(2)加热明显增强Casepase-3的表达和降低Survivin的表达,从而促进肺癌细胞的凋亡。(3)加热温度42℃、加热时间2h为肺癌细胞热疗的最佳条件。
Objective:To observe the effects of hyperthermia on apoptosis,cellular necrosis and expression of survivin and casepase-3 gene of lung cancer A549 cell,and to investigate its mechanisms.Methods:Lung cancer cell A549 at exponential phase of growth was heated at the same temperature for different time and for the same time at different temperature. After this dealing, to detect cell apoptosis and cellular necrosis condition by flow eytometry (FCM). To detecting apoptosis-related gene expression of casepase-3 and survivin by RT-PCR. Results:Cell apoptosis increased after hyperthermia. And that at 42℃ and 2h heating was the obvious one. With the temperature and time increased in hyperthermia,the cellular necrosis increased at the same time. Hyperthermia could enhance expression of casepase-3 and reduce expression of survivin significantly. 42℃ 2h heating was the best result of all.Concluslon: Hyperthermia can enhance expression of casepase-3 and reduce expression of surviviu obviously, and then encourage apoptosis of cancer cell. The 42℃ 2h is the best one. Heating for 2h at 42 ℃ is the optimum condition for hyperthermia.
