山仙颗粒含药血清对大鼠S-180肉瘤细胞fas基因表达的相关性研究 被引量:1

摘要 目的:探讨山仙颗粒(SXG)含药血清对大鼠S-180肉瘤的抑制作用及肿瘤凋亡相关基因fas表达的影响,探讨其抗肿瘤发生和发展的机理。方法:通过体外细胞培养扩增S-180肉瘤细胞制备细胞悬液,将其置入不同浓度的含药SD大鼠血清及正常SD大鼠血清中,分成对照组(1640培养液)、空白1组(10%正常鼠血清)、空白2组(100%正常鼠血清)、含药血清1组(10%鼠含药血清)和含药血清2组(100%鼠含药血清),分别培养12h、24h、48h后收集细胞制成细胞涂片,免疫细胞化学(二步法)检测瘤组织中fas基因的表达情况对照分析。结果:与对照组及空白1、2组比较,鼠含药血清2组fas基因表达水平升高(P<0.05),对照组、空白1、2组及鼠含药血清1组无显著差异(P>0.05)。结论:SXG大鼠含药血清能显著使体外培养的S-180肉瘤的促凋亡基因fas表达增强,从而诱导S-180肉瘤细胞的凋亡来抑制其生长。
