摘要
目的探索IL-15对CIK细胞的扩增及细胞表型的影响。方法以常规培养的CIK细胞为对照,观察培养体系中加入IL-15后CIK细胞的扩增情况,并用流式细胞仪检测其表面标志的变化。结果①IL-15组的扩增速度大于常规培养组,从培养14天起两组扩增倍数的差异有统计学意义(P<0.05);②培养后IL-15组的CD3+ CD56+细胞比例及NKG2D表达率均高于常规培养组,于培养14天起,两组差异均有统计学意义(P<0.05)。结论 IL-15可增加CIK细胞的增生能力,并增加CD3+ CD56+细胞及NKD2D的表达率。
ObjectiveTo investigate the effect of interleukin-15 (IL-15) on the proliferation of cytokine-induced killers (CIKs) and on the expression of cell surface markers.MethodsMononuclear cells were isolated from peripheral blood of 15 healthy volunteers by Ficoll density gradient centrifugation, and then were induced to differentiating into CIKs in the presence of various cytokines with or without IL-15. Cells were harvested each week for 4 weeks starting with day 0, and viable cell number was determined using the Trypan blue dye-exclusion method. The levels of cell surface markers CD3, CD56, and NKG2D were measured by flow cytometry. ResultsCIKs in the IL-15-treated group had a significantly higher proliferation rate from day 14 than those in the group without IL-15 treatment (P〈0.05). Moreover, IL-15 treatment resulted in a significantly higher proportion of CD3+CD56+ cells as well as enhanced expression of NKG2D as compared to the IL-15-untreated group (P〈0.05).ConclusionIL-15 increases the proliferation of CIKs and enhances the expression of NKG2D on the surface of CD3+CD56+CIKs.
