反相HPLC法测定鼠血浆中甲基强的松龙及其前体酯

摘要 为建立测定血浆中甲基强的松龙(MP)及其前体酯甲基强的松龙琥珀酸酯(MPHS)的高效液相色谱(HPLC)法。采用HPLC法检测大鼠静脉注射甲基强的松龙琥珀酸钠(MPS)后不同时间血浆中MP及MPHS的浓度。采用YWG—C_18分析柱,以乙腈∶水∶醋酸—醋酸钠缓冲液(29∶71∶1.4,V/V/V)为流动相,在254nm波长下监测洗提液。结果标准曲线的相关系数大于0.9993,日内及日间变异系数分别为(1.9~5.6)%和(4.1~14.7)%。结果提示本法系国内首创,具有操作简便、快速、灵敏、准确等特点。 Establish the method of High Performance Liquid Chromatographic(HPLC) for determination of Methylprednisolone (MP)and its ester prodrug Methylprednisolone Hemisuccinate (MPHS) in rat plasma. Using HPLC to detect the plasma concentration of MP and MPHS at different time after intravenous injection Methylprednisolone Sodium Succinate (MPS) in SD rats. Reversed -phase liquid Chromatography was performed on a YWG-C18 column(sphersirb 10μm)using a mobile phase of acetonitrile-water-acetate buffer(29 : 71: 1. 4, V/V/V)with ultraviolet detection (254nm). The coefficient of correlation of calibration curves is more than 0. 9993. CV of Intra and inter assay for plasma samples is(l. 9-5. 6)% and (4.1-14. 7)%,respectively. The assay methodology is the only one in which MP and MPHS are measured simultaneously in plasma in China and is simple. rapid,sensitive and accurate for the quantitative determination of MP and MPHS in SD rat plasma.
