天麻钩藤饮对血管紧张素Ⅱ诱导血管平滑肌细胞A7r5增殖的影响

Inhibitory Effects of Tianma Gouteng Decoction on Rats Vascular Smooth Muscle Cells A7r5 Proliferation Induced by Angiotensin Ⅱ

目的观察天麻钩藤饮对血管紧张素Ⅱ(AngⅡ)诱导血管平滑肌细胞(A7r5)增殖的影响及机制。方法 AngII刺激A7r5细胞建立细胞增殖模型。采用CCK-8法检测细胞增殖,观察天麻钩藤饮(0.25,0.5,1,2 mg.mL-1)、亚硝基-精氨酸甲酯(100μmo1.L-1)对AngII诱导血管平滑肌细胞增殖的影响。硝酸还原酶及化学比色法测定细胞上清液中一氧化氮(NO)、一氧化氮合酶(NOS)、诱导型一氧化氮合酶(iNOS)水平。结果天麻钩藤饮在0.25～2 mg.mL-1呈剂量及时间依赖性抑制A7r5增殖;天麻钩藤饮能升高NO、NOS、iNOS水平。结论天麻钩藤饮对AngII诱导的A7r5细胞增殖有抑制作用,升高NO和NOS水平可能是其发挥作用的机制。

Objective To investigate effects of Tianma Gouteng Decoction（TGD） on rats vascular smooth muscle cells ATr5 proliferation induced by angiotensin II（Ang Ⅱ）, and its mechanism. Methods Cell proliferation model was established by stimulation with AngⅡ. Cell proliferation was measured by CCK-8 assay. Effects of TGD （0.25, 0.5, 1 and 2 mg·mL^-1） and N-nitro-L-arginine methyl ester （L-NAME, 100 μmol ·L^-1） on AngⅡ-induced cell proliferation were observed. Nitric oxide（NO） level was measured by Griess reagent. Nitric oxide synthase（NOS） and inducible nitric oxide synthase （iNOS） levels were detected by chemical colorimetrie method. Results TGD at the dose range of 0.25 mg·mL^-1 to 2 mg·mL^-1 inhibited cell proliferation in a dose and time dependent manner. TGD markedly increased NO, NOS and iNOS levels. Conclusion TGD exerts an inhibition of A7r5 cell proliferation induced by AngⅡ, and its mechanism might be associated with the increase of the secretion of NO and NOS.
