摘要
目的比较SD、Wistar和F344三种品系大鼠对天花粉蛋白诱导的被动皮肤过敏(Passive cutaneous ana-phylaxis,PCA)反应的差异性。方法选用SD、Wistar和F344三种品系大鼠,用天花粉蛋白为致敏原,以PCA反应时出现蓝斑的直径及蓝斑OD值变化率作为判别标准,进行大鼠同种或小鼠异种PCA试验。结果大鼠同种PCA试验显示,每只大鼠注射天花粉蛋白0.9 mg后,三种品系大鼠PCA反应均为阳性,蓝斑直径大小及蓝斑OD值变化率从大到小依次为:SD、F344大鼠>Wistar大鼠;小鼠异种PCA试验显示,耳廓皮内分别注射三种品系大鼠天花粉蛋白抗血清后,小鼠异种PCA反应均为阳性,耳廓蓝染OD值变化率从大到小依次为:SD大鼠抗血清组>F344大鼠抗血清组>Wistar大鼠抗血清组。结论三种品系大鼠对天花粉蛋白的敏感性存在差异,其中以SD大鼠最为敏感。
Objective To compare the variability of passive cutaneous anaphylaxis (PCA) caused by trichosanthin (TCS) among Sprague Dawley(SD), Wistar and Fischer344(F344) rats. Methods Homologous PCA was performed on SD, Wistar and F344 rats, and xenogenic PCA was performed on KM mice. TCS was taken as the allergen. The diameter of dermal Evan's blue spot and the optical density were measured to evaluate the severity of PCA. Results The results of homologous PCA showed that PCS was positive in all of the three rat strains after injection of 0.9mg TCS for each rat. According to the the diameter of Evan's blue spots and the optical density, PCA was stronger in SD and F344 rat than that in Wistar rat. Xenogeneie PCA was positive in KM mice after intradermally auricular injection of TCS antiserum of the three different rat strains. According to the optical density of Evan's blue spots, the intensity of antiserum was as follows: SD rat antiserum〉 F344 rat antiserum 〉 Wistar rat antiserum. Conclusion There are variabilities in sensitivity of TCS-induced PCS among the three rat strains, and SD rat is the most sensitive one.
