菘蓝有效成分抗流感病毒作用的实验研究 被引量:7

摘要 目的研究菘蓝有效成分在体内外抗甲型流感病毒的作用。方法体外用MTT法检测菘蓝有效成分对传代狗肾细胞(MDCK)的毒性浓度,观察菘蓝有效成分抑制甲型流感病毒的致细胞病变作用(CPE),MTT法检测菘蓝有效成分对流感病毒的抑制率;建立在体流感病毒感染小鼠肺炎模型,通过观察小鼠肺病变程度指标,测定肺组织内流感病毒血凝滴度判定菘蓝有效成分对小鼠肺炎的治疗作用。结果菘蓝有效成分在体外主要抑制甲型流感病毒在细胞内的复制,其对病毒的半数抑制率浓度为64μg.mL-1,治疗指数达28.88;口服菘蓝有效成分能明显减轻小鼠肺组织病变程度,对肺组织内病毒的增殖有明显抑制作用。结论菘蓝有效成分具有较好的抗甲型流感病毒的作用,且呈剂量效应关系。 Objective To investigate the antiviral effect of active component of Isatis tinctoria L.(ITL) on influenza virus A/H1N1 in vitro and in vivo. Methods Methyl thiazolyl tetrazolium(MTT) assay was used for the detection of irt-vitro toxicity concentrations of the ITL active component on Madin-Darby canine kidney(MDCK) cell line. Then cytopathic effect(CPE) of ITL active component on influenza virus A/H1N1 was observed, and MTF assay was applied to observe the inhibitory rate for influenza virus. Influenza virus infection mouse pneumonitis model was established, and pulmonary pathological changes and pulmonary virus hemagglutination titers of the mice were examined for the assessment of the anti-lnfluenza activity of the active component in vivo. Results The activity component had inhibitory effect on viral replication in vitro with IC50 being 64 μg·mL^-1 and therapeutic index(TI) up to 28.88. Oral use of active component significantly reduced the pulmonary pathological changes, and inhibited the proliferation of pulmonary virus.Conclusion The active component of Isatis tinctoria L. has antiviral activity by inhibiting replication of influenza virus A/H1N1 in vitro and in vivo in a dose-dependent manner.
