黄芩中总黄酮的提取及高速逆流色谱分离纯化 被引量:3

Total Flavonoids Extraction from Radix Scutellariae and Its Purification by High-speed Counter-current Chromatography

摘要 目的探讨中药黄芩中总黄酮的提取方法,并应用高速逆流色谱进行分离纯化,得到纯度较高的总黄酮有效部位。方法采用正交设计法乙醇回流提取得到总黄酮粗提物,再应用高速逆流色谱进行分离纯化。结果最佳提取工艺条件为:60%乙醇提取2次,溶剂量分别为12倍、10倍,时间分别为2 h、1 h,粗提物总黄酮含量达15.8%;高速逆流色谱两相溶剂系统为氯仿-甲醇-水-醋酸(4∶3∶3∶0.1,V/V),进样量为400 mg,得到45 mg纯度达90.2%的总黄酮有效部位。结论本实验应用乙醇回流提取及高速逆流色谱分离纯化可得到纯度较高的黄芩总黄酮有效部位。 Objective Total flavonoids extraction process h'om Radix Scutellariae by ethanol reflux was investigated, and high-speed counter-current chromatography( HSCCC )was applied for the separation and purification of flavonoids. Methods The optimal extraction condition for flavonoids was investigated with orthogonal test design, and high-purity target of flavonoids was obtained by HSCCC. Results The optimal condition of extraction was: extraction with 60 % ethanol for two times, the solvent being 12, 10 times as much as the herbal materiall, and extraction time being 2 hours, 1 hour. Under the above condition, content of total flavonoids in the crude extract was up to 15.8 %. The experiment of HSCCC was performed with a two-phase solvent system composed of chloroform-methanol-water-acetic acid (4 : 3 : 3 : 0.1, V/V). When the injection volume of crude extract was 400 rag, 45 mg total flavonoids was obtained with a purity of 90.2 %. Conclusion High-purity flavonoids from Radix Scutellariae is obtained after ethanol reflux extraction and purification by HSCCC.
