摘要
目的:为了解TNFαMAb对大鼠AHNP时ALI是否具有防治作用。方法:利用AHNP并发ALI大鼠模型,将实验大鼠随机分为三组:假手术组、AHNP组和TNFαMAb预处理组,动态观察大鼠24小时存活情况、PaO2和PaCO2、肺组织MPO活性变化、肺含水量改变以及胰、肺组织的病理改变。结果:TNFαMAb尽管没有明显改变胰腺组织的病变,但能明显降低AHNP大鼠24小时死亡率(TNFαMAb预处理组10%,AHNP组50%,P<0.05),减少肺脏PMN的聚集,减轻肺组织的损伤程度。结论:TNFα是AHNP引起ALI发生的重要炎症介质,TNFαMAb通过阻断TNFα的作用和减少肺内PMN的聚集,对AHNP时肺组织起保护作用。
Objectives: To study the effects of tumor necrosis factor Q monoclonal antibody (TNF.MAb) on acute lung injury (ALI) induced by acute hemorrhagic and necrotic pancreatitis (AHNP) in rats. Methods: Adopting an model of rat AHNP-induced ALI, experimental rats were di-vided into three groups: control group, AHNP group and TNF.MAb group. The changes of 24-hour-rat death rate, PaO2 and PaCO2, pulmonary myeloperoxidase (MPO) activity, lung water contents and pathological changes in lung were examined. Results: Although TNF.MAb didn,t change the injury de-gree of pancreas, TNF.MAb significantly decreased the 24-hour-rat death rats (TNF.MAb group:10%, AHNP group: 50%,P<0.05), reduced the pulmonary polymorphonuclear neutrophil (PMN),improved the pulmonary injury degrees. Conclusions: TNF. is an important inflammatory medium for AHNP-induced ALI- TNF.MAb can neutralize TNF. activity and reduce the PMN accumulation in lung tissue, which preserve the lung tissue with AHNP.
