虎杖对α-葡萄糖苷酶的抑制作用 被引量:3

摘要 [目的]筛选虎杖(Polygonum cuspidatum Sieb.et Zucc.)α-葡萄糖苷酶抑制活性物质。[方法]对中药虎杖的α-葡萄糖苷酶抑制活性进行测定,并对其活性物质进行了初步分离。[结果]虎杖水提物浓度为50 mg(生药)/ml时,对α-葡萄糖苷酶活性的抑制率达68.5%,IC50约为37 mg/ml。虎杖水提物是α-葡萄糖苷酶的非竞争性抑制剂,抑制常数Ki为33 mg/L。采用层析分离得到了虎杖α-葡萄糖苷酶抑制剂,初步鉴定为多糖物质。[结论]该研究为植物源α-葡萄糖苷酶抑制剂的开发奠定了基础,对发现高效、安全的降低餐后血糖新药物具有重要作用。 [Objective]The research aimed to screen α-glucosidase inhibitor from Polygonum cuspidatum Sieb.et Zucc.[Method] The ability of the extract from Polygonum cuspidatum Sieb.et Zucc.to inhibit α-glucosidase was studied and the crude inhibitor was screened.[Result]The results showed that the inhibition rate of 50 mg/ml extract to α-glucosidase was 68.5%,its IC50 was about 37 mg/ml.The Polygonum cuspidatum Sieb.et Zucc.extract was noncompetitive inhibitior of α-glucosidase,and the Ki was 33 mg/L.The crude inhibitor purified through DEAE-cellulose DE-52 was identified as polysaccharides.[Conclusion] The study can lay basis for developing botanical α-glucosidase inhibitor,and has important effect on finding new reducing postprandial plasma glucose medicine of high active and safety.
