轻度认知功能障碍及老年性痴呆患者外周淋巴细胞糖原合成酶激酶-3的变化

Glycogen synthase kinase-3 in the lymphocytes of patients with mild cognitive impairment and Alzheimer's disease

摘要 目的探讨轻度认知功能障碍(MCI)及阿尔茨海默病(AD)患者外周淋巴细胞糖原合成酶激酶-3(GSK-3)的变化。方法用放射性配体结合试验检测正常老年人(对照组)、MCI患者(MCI组)及AD患者(AD组)外周淋巴细胞GSK-3的活性,并用Westernblot法检测GSK-3pSet9位点的磷酸化水平及总GSK-3B表达。结果AD组及MCI组GSK-3活性分别为(1.32±0.09)和(1.22±0.10),明显高于对照组的(1.00±0.06),差异有统计学意义(均P〈0.01);AD组及MCI组GSK-3BSer9位点的磷酸化水平分别为(0.82±0.07)和(0.87±0.08),明显低于对照组的(1.01±0.07),差异有统计学意义(P〈0.05或0.01);AD组及MCI组总GSK-3p表达水平与对照组比较,差异无统计学意义(P〉0.05)。结论MCI及AD患者外周淋巴细胞GSK-3的活性升高,可能对此类疾病的早期诊断有潜在的价值。 Objective To explore changes in glycogen synthase kinase-3 (GSK-3) levels in the lympho- cytes of patients with mild cognition impairment (MCI) and Alzheimer's disease (AD). Methods GSK-3 activity was measured by 3219 liquid scintillography in a control group, an MCI group and an AD group. The expressions of GSK-313at Ser9 and total GSK-313 were determined by western blotting. Results GSK-3 levels were significantly higher in the AD and MCI groups than in the control group. The phosphorylation of GSK-313at Ser9 was significantly lower in the AD and MCI groups compared with the control group, but there was no difference in the phosphorylation of total GSK-313 among the three groups. Conclusions Higher GSK-3 activity in the lymphoeytes might have value in the diagnosis of MCI and early AD.
