不同产地黄芪中黄芪皂苷Ⅰ、黄芪皂苷Ⅱ和黄芪皂苷Ⅳ含量测定 被引量:25

Determination of astragaloside Ⅰ,astragaloside Ⅱ and astragaloside Ⅳ in Radix Astragali from various habitats

摘要 目的:研究采用HPLC-ELSD法测定不同产地黄芪药材中的黄芪皂苷Ⅰ、黄芪皂苷Ⅱ和黄芪皂苷Ⅳ的含量。方法:色谱柱为ZORBAX Eclipse XDB-C18(250 mm×4.6 mm,5μm),流动相为乙腈和水,梯度洗脱,流速1 mL·min^-1,柱温35℃;蒸发光散射检测器,ELSD参数:喷雾器温度为39℃,漂移管温度55℃,载气压力172.4 kPa,增益值为200。结果:黄芪皂苷Ⅰ、黄芪皂苷Ⅱ和黄芪皂苷Ⅳ的线性范围分别为0.8312~8.312μg(r=0.9996),2.544~16.536μg(r=0.9990),0.328~3.28μg(r=0.9992);平均回收率分别为99.1%,98.0%,98.6%,RSD分别为3.9%,2.8%,2.8%。结论:该方法简单快捷,适合于黄芪药材中黄芪皂苷的含量测定研究,为寻找更佳黄芪产地提供了依据。 Objective:To establish an HPLC-ELSD method for the determination of the content of astragaloside Ⅰ,astragaloside Ⅱ and astragaloside Ⅳ in Radix Astragali from various habitats.Methods:The conditions of chromatography:ZORBAX Eclipse XDB-C18(250 mm×4.6 mm,5 μm)was used with a mobile phase of CH3CN-H2O,flow rate:1 mL·min^-1,column temperature:35 ℃,:spray temperature 39 ℃,drift tube temperature:55 ℃,air pressure:172.4 kPa,gain:200.Results:The calibration curves of astragaloside Ⅰ,astragaloside Ⅱ and astragaloside Ⅳ were in good linearity over the ranges of 0.8312–8.312 μg(r=0.9996),2.544–16.536 μg(r=0.9990) and 0.328–3.28 μg(r=0.9992) respectively;The average recoveries were 99.1%,98.0% and 98.6% with RSD of 3.9%,2.8% and 2.8%,respectively.Conclusion:The method is simple,rapid,sensitive and reproducible for determination of astragalosides in Radix Astragali.and can be used a basis for finding better habitat of Radix Astragali.
