摘要
目的:检测三磷酸腺苷结合盒转运体A3(ABCA3)及P-糖蛋白(P-gp)基因在急性白血病(AL)细胞中的表达水平,并分析2种基因表达的临床意义。方法:应用RT-PCR方法检测42例初治、21例复发/难治成人AL患者及10例正常对照者骨髓单个核细胞ABCA3和P-gpmRNA表达水平。结果:初治AL组ABCA3、P-gp的表达水平均高于正常对照组(均P<0.01);复发难治AL组2种基因的表达水平均明显高于初治组(均P<0.01);急性淋巴细胞白血病(ALL)组ABCA3、P-gp的表达水平均高于急性髓细胞白血病(AML)组(均P<0.05)。AL患者ABCA3、P-gp表达水平呈正相关(r=0.599,P<0.01)。初治AL患者中获完全缓解组AB-CA3、P-gp的表达水平均低于未完全缓解组(均P<0.05)。结论:ABCA3、P-gp与AL患者的多药耐药及治疗预后密切相关,且2种基因的表达水平呈正相关。
Objective:To investigate the expressions of ATP binding cassette transporter A3(ABCA3)and P glycoprotein(Pgp)gene in acute leukemia(AL)patients and to analyze the clinical significance of the two genes' expressions.Method:Bone marrow mononuclear cells from 42 newly diagnosed,21 relapse/refractory AL patients and 10 normal controls were analyzed.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the expressions of ABCA3 and Pgp.Result:The levels of ABCA3 and Pgp gene in the newly diagnosed AL groups were higher than those in normal controls(P0.01);The two gene's expressions in the relapse/refractory AL groups were significantly higher than those in the newly diagnosed groups(P0.01).The levels of ABCA3 and Pgp gene expression in acute lymphocytic leukemia(ALL)groups were higher than those in acute myeloid leukemia(AML)groups(P0.05).The expressions of the two genes were positively correlated(r=0.599,P0.01).Compared to noncomplete remission group,the complete remission group had lower expressions of ABCA3 and Pgp.Conclusion:The expressions of ABCA3 and Pgp were closely related to the multidrug resistance and bad prognosis of AL,and there was a significant positive correlation between Pgp and ABCA3 expressions in AL.
