摘要
目的:探讨慢性再生障碍性贫血患者(cAA)血浆及CD3+、CD8+细胞胞质内干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的表达情况及其在cAA发病机制中的作用。方法:应用ELISA法检测50例cAA患者及50例健康志愿者血浆IFN-γ、TNF-α的水平,流式细胞仪(FCM)检测外周血CD3+、CD8+细胞胞质内IFN-γ、TNF-α的表达水平。结果:cAA患者血浆中IFN-γ、TNF-α的水平显著升高,与正常对照组相比,差异有统计学意义(P<0.05),CD3+、CD8+细胞胞质内IFN-γ、TNF-α的水平亦显著升高,与正常对照组相比,差异有统计学意义(P<0.05)。结论:cAA患者T淋巴细胞高表达IFN-γ、TNF-α,其中CD8+T淋巴细胞亦高表达IFN-γ、TNF-α,这可能是cAA骨髓造血功能衰竭的机制之一。
Objective:To investigate the expression of interferon-gamma(IFN-γ)and tumor necrosis factor-α(TNF-α)in peripheral blood of patients with chronic aplastic anemia(cAA)and determine the role of these cytokines in the pathogenesis of cAA.Method:The expressions of IFN-γ and TNF-α were detected in blood plasma by ELISA and in CD3+,CD8+ T lymphocytes by flow cytometry from 50 cAA patients and 50 healthy volunteers.Result:Elevated levels of IFN-γ and TNF-α were found in blood plasma and CD3+,CD8+ T lymphocytes in AA patients compared with healthy controls(P0.05).Conclusion:The overexpression of IFN γ and TNF-α in T cells,including CD8+ T cells,may be one of the mechanisms for bone marrow failure in cAA patients.
