肝癌组织蛋白酶B及其基因表达与临床病理因素的关系

Expressions and effects of cathepsin B and its mRNA in hepatocellular carcinoma and their clinicopathological significance

摘要 [目的]探讨肝癌(HCC)组织中组织蛋白酶B(CB)和CB mRNA的表达和作用。[方法]应用免疫组织化学和RT-PCR方法测定71例肝组织标本CB及CB mRNA,其中对照(C)组12例,HCC组34例和HCC癌旁组织(PTT)组25例,对比分析CB、CB mRNA与肝癌临床病理因素的关系。[结果]HCC组CB的平均吸光度值和CBmRNA条带的光密度比值均明显高于C组和PTT组(均P<0.01),PTT组与C组间均无显著差异(P>0.05)。CB在包膜不完整,有肝内转移和肝外转移HCC中的表达分别明显高于包膜完整,无肝内转移和肝外转移者(P<0.05或<0.01);与肿瘤的大小无关(P>0.05);随组织分化降低,均有增高趋势,但无统计学意义。[结论]HCC组织CB及其基因均呈现高表达,可能在HCC的发生、发展,尤其是侵袭和转移过程中发挥重要的作用。 [Objective]To study the expressions and effects of cathepsin B(CB)and its mRNAin hepatic carcinoma.[Methods]Expressions of CB and its mRNA in 71 specimens of human hepatic tissue,including 12 of C group,34 of HCC group and 25 of PTT group were detected with immunohistochemistry and RT-PCR.Relationships between the expression of CB and its mRNA and clinicopathological factors were analysed.[Results]Expressions of CB protein and its mRNA in HCC group were significantly higher compared with C group or PTT group(all P0.01),there was no difference between PTT group and C group(all P0.05).Expressions of CB protein and its mRNA in HCC with invaded capsule,intrahepatic and extrahepatic metastasis,were significantly higher than those in HCC with integral capsule,without intrahepatic and extrahepatic metastasis(P0.05 or P0.01),these expressions were not significantly related to the size of HCC(all P0.05).With lower Edmondson grade,expressions increased of no statistical significance.[Conclusion]The CB and its mRNA present high expressions in HCC group and they may play an important part in the generation,development,invasion and metastasis of HCC.
