摘要
目的:为阐明牙齿发育的机制,探讨程序性细胞死亡(PCD)及其相关调控基因在牙齿发育中的作用。方法:利用原位末端标记法、原位杂交及免疫组化技术对SD大鼠牙齿发育不同时期PCD及Bcl-2、Bax基因的表达进行研究。结果:在牙胚发育早期的生长中心处PCD、Bcl-2、BaxmRNA及其蛋白的表达均很强,牙体组织形成后PCD及Bcl-2mRNA蛋白的表达逐渐减弱,但Bax蛋白的表达仍然明显。结论:Bcl-2。
Objective: To investigate the role of programmed cell death (PCD) and its regulating genes in tooth development. Methods: PCD and the expression of both transcription and translation of Bcl 2 and Bax genes in different stages of tooth development of SD rat were observed by Tdt mediated dUTP nick end labeling (TUNEL), mRNA in situ hybridization and immunohistochemical staining. Results: The number of PCD cells and the expression of Bcl 2 and Bax predominantly appeared at growth initiation center in the earlier stage of tooth germ, while the expression of PCD and Bcl 2 gradually decreased following dentin formation with strong expression of Bax . Conclusion: Bcl 2 and Bax genes and PCD are important regulating factors in tooth development.
