摘要
目的:评价姜黄素对人单核细胞株THP-1中炎症相关因子NF-κB、TNF-α和IL-1βmR-NA表达的影响。方法:采用佛波酯(TPA)构建鼠耳肿胀急性炎症反应模型,采用不同浓度的姜黄素处理THP-1细胞,1 μg/mL脂多糖(LPS)刺激细胞4 h,提取细胞总RNA。用RT-PCR方法检测NF-κB、TNF-α和IL-1β mRNA的表达。结果:局部外用0.8和1.6 μmol的姜黄素及腹腔注射50 mg/kg和100 mg/kg姜黄素,对鼠耳肿胀急性炎症反应均有明显抑制作用。浓度为50 μg/mL和25μg/mL的姜黄素可以明显抑制THP-1细胞中LPS诱导的NF-κB、TNF-α和IL-1β mRNA的表达。结论:姜黄素对炎症因子有抑制作用。
Objective: To assess the effects of curcumin on the expression of inflammatory cytokines in THP - 1 cells in vivo. Methods: An ear inflammation model was established by 12 - o- tetradecanoylphorbol - 13 - acetate (TPA) in mice and THP- 1 cells treated with different concentration of curcumin. After the treated THP- 1 ceils were stimulated with 1 μg/mL lipopolysaccharide (LPS) for 4 hs, total RNA was abstracted. The expression of NF- κB, TNF - α and IL - 1β was detected by RT - PCR. Results: Topical application of 0.8μmol and 1.6 μmol or intra -abdomenal injection of 50 mg/kg and 100 mg/kg of curcumin markedly inhibited TPA - induced epidermal inflammation of mice ears. Curcumin at the concentrations of 50 μg/mL and 25μg/mL significantly inhibited the expression of NF- κB, TNF -α and IL- 1β mRNA. Conclusion: Curcumin can inhibit inflammatory reaction.
