摘要
目的:探索茶多酚对肺鳞癌细胞的抑制效应及相关机制。方法:以肺鳞癌PC10为研究对象,进行裸鼠成瘤实验。观察茶多酚对裸鼠成瘤的影响。应用免疫组织化学方法检测细胞凋亡相关Bcl2和Caspase3和增殖相关蛋白PCNA的表达,应用TUNNEL法检测细胞凋亡。结果:茶多酚可抑制裸鼠移植瘤的生长。茶多酚可促进促凋亡蛋白caspase3表达并抑制抑凋亡蛋白Bcl2表达,并抑制增殖细胞核抗原的表达。TUNNEL实验结果提示茶多酚可促进PC10细胞凋亡。结论:茶多酚可抑制肺鳞癌移植瘤生长,与促进癌细胞凋亡和抑制细胞增殖有关。
Objective:To investigate the inhibitory effect and related mechanism of EGCG on transplantation tumor from lung squamous cell carcinoma cell line PC10.Methods:lung squamous cell carcinoma cell line PC10 transplantation tumor model was established and effect of EGCG on growth of transplantation tumor were detected.Cell apoptosis and the expression of proliferation associated protein were detected by immunohistochemical mothod.Cell apoptosis was detected by TUNNEL method.Results:EGCG could inhibit tumor growth in nude mice.The expression of apoptosis-promoting protein caspase3 was enhanced while the expression of inhibiting-apoptosis protein Bcl2 and proliferation associated protein PCNA were inhibited in EGCG group compared with control group.TUNNEL results showed that EGCG accelerated cell apoptosis Conclusions:EGCG can inhibit the growth of PC10 cell transplantation tumor in nude mice which may have relationship with inhibition of proliferation and enhancement of cell apoptosis.
