摘要
目的:观察栀子浸膏对Hulth法复制兔膝骨关节炎模型中软骨基质的改变及软骨与滑膜中IL-1的影响。方法:36只大耳白兔随机分为给药组、模型组和对照组,每组12只。给药组、模型组行Hulth术法复制骨关节炎模型,术后第5周,给药组以栀子浸膏600mg/kg灌胃,模型组与对照组给予等量生理盐水灌胃,给药3周后解剖显微镜下行软骨PelletierJP评分,取股骨内侧髁关节软骨行Masson及甲苯胺蓝染色观察软骨中胶原纤维、蛋白多糖含量的变化,免疫组化检测软骨与滑膜中IL-1的表达情况。结果:栀子浸膏能延缓骨关节炎关节软骨基质的丢失,给药组软骨与滑膜中IL-1的表达明显低于模型组,P<0.05。结论:栀子浸膏可减少骨关节炎模型软骨与滑膜中IL-1的表达和保护关节软骨基质,从而延缓病程的进展。
Objective:To observe the change of the Gardenia Jasminoides Ellis extract to the articular cartilage matrix and the effect of IL-1 in cartilage and synovial on rabbit knees osteoarthritis models by Hulth method. Methods: 36 rabbits were randomly divided into three groups: administration group (n=12), model group (n=12), control group (n=12). administration group and model group were duplicated the osteoarthritis model by Hulth method. after 5 weeks, the administration group were administered intragastrically with 600mg/kg of the extract of Gardenia Jasminoides Ellis, the model and the control group were administered intragastrically with equivalent normal saline. After 3 weeks medication, the cartilage been scored by Pelletier JP method under the dissecting microscope. the articular cartilage of the medial femoral condyle was Masson staining and Toluidine Blue staining. After that, observe the change of collagen fiber and proteoglycan in cartilage and detect IL-1 expression in cartilage and synovial by immunohistochemical technique. Results: The Gardenia Jasminoides Ellis extract could delay the lose of the articular cartilage matrix in osteoarthritis, The IL-1 expression in cartilage and synovial of administration group is lower significantly than the model group, P0.05. Conclusion: The Gardenia Jasminoides Ellis extract could reduce the IL-1 expression in cartilage and synovial and protect the articular cartilage matrix from lost in osteoarthritis model, and slow down the process of the osteoarthritis.
