大孔树脂法纯化牛蒡子中有效部位的工艺 被引量:5

摘要 目的建立使用大孔树脂对牛蒡子提取液进行纯化的可靠方法。方法以牛蒡苷含量为指标,采用HPLC法测定其含量。考察大孔树脂对其的吸附性能和洗脱参数,主要包括:树脂的上样量、上样液pH、树脂径高比、流速、洗脱液体积以及浓度等参数,同时考察了树脂的重复使用次数及再生后吸附量的变化。结果最佳工艺为:用HPD100大孔吸附树脂,树脂用量为药材的3倍,调节上样液pH值为7,树脂径高比为:1∶10,1.5 BV/h流速上样,洗脱液采用40%乙醇,洗脱流速为0.7 BV/h。该树脂重复使用3次后,需要进行再生,再生后还可继续使用。结论该方法具有可行性,符合研究要求。 Objective To establish a reliable method for purifying the extraction from fructus arctii by macroporous resin. Methods With the content of arctiin as criteria,the content of it was determined by using HPLC and investigated the adsorption performance and the elution parameters,including: the amount of resin,the pH of sample for loading,the ratio of dia-height,flow rate,flush volume and the concentration of eluent,were investigated.The times for repeated use and recovery capacityafter regeneration were studied. Results The optimized purification methods were summarized as follows: the capacity of HPD100 macroporous adsorptive resin was as much as 3 times of the sample load,the pH of sample for loading was 7,the flow rate of sample was 1.5 BV/h,and 70% ethanol was used as eluant solution in the flow rate of 0.7 BV/h.The resin could be reused three times,and need for regeneration,the resin after regeneration can be used again. Conclusions The established method is reliable and efficient for the purification of extraction from fructus arctii.
