二苯乙烯苷对SAMP8鼠脑组织氨基酸类神经递质的影响 被引量:5

摘要 目的观察二苯乙烯苷(TSG)对快速老化小白鼠/8(SAMP8)脑组织氨基酸类神经递质含量的影响。方法选用6月龄的SAMP8鼠50只,随机分为5组,每组10只:SAMP8对照组,阳性药(石杉碱甲)对照组,TSG低、中、高剂量组,另选用6月龄SAMR1鼠10只作正常对照组。各组分别灌胃相应药物50天后,采用高效液相色谱法检测各组小鼠脑组织氨基酸类神经递质含量。结果与正常对照组SAMR1组比较,SAMP8对照组脑组织谷氨酸(Glu)、谷氨酰胺(Gln)、天冬氨酸(Asp)、γ-氨基丁酸(GABA)含量明显升高,差异有显著性(P<0.05或P<0.01),天冬酰胺(Asn)、甘氨酸(Gly)有升高趋势,但差异无显著性(P>0.05);TSG各剂量组能显著降低SAMP8鼠Glu、Asp含量(P<0.05或P<0.01),但对Gly的降低作用不明显(P>0.05)。部分TSG剂量组对降低Gln、Asn和GABA含量作用明显(P均<0.05)。结论 TSG能显著降低SAMP8鼠脑组织Glu、Asp含量,纠正了SAMP8鼠脑区氨基酸类递质代谢的紊乱状态,使兴奋性和抑制性氨基酸神经递质最终趋向平衡。 Objective To observe the effects of 2, 3, 5, 4' - tetrahydroxy - stilbene - 2 - O - 13 - D - glyco- side (TSG) on the contents of amino acids neurotransmitter of senescence accelerated mouse prone/8 (SAMP8) mice brain tissues. Methods 50 SAMP8 mice aged 6 months were selected in this study and were randomly divided into 5 experimental groups: SAMP8 untreated control group, huperzine A control group, low - , mid - and high - dose groups of TSG, each group had 10 mice. Another 10 senescence accelerated- resistant mouse/1 (SAMR1) mice aged 6 months were selected as normal control group. After each group were infused orally with corresponding drugs for 50 days continuously, the contents of amino acids neurotransmitter in brains were detected by using HPLC. Resuits SAMP8 group yielded the elevated levels of Glu, Gln, Asp and GABA of the brain tissues, compared with SAMR1 group, there were statistically significant differences ( P 〈 0.05 or P 〈 0.01), and the contents of Ash and Gly increased slightly, there were no statistical differences ( P 〉0.05). In TSG- treated groups, the contents of Glu and Asp decreased significantly, compared with SAMP8 group, there were statistical differences ( P 〈 0.05 or P 〈0.01), but the contents of Gly decreased slightly ( P 〉0.05). Some doses of TSG could down- regulate Gln, Asn and GABA ( P 〈 0.05). Conclusion TSG can obviously down - regulate Glu and Asp, and can retrieve the metabolic disorder of amino acids neurotransmitter in SAMP8 mice brain tissues, resulting in the balance of the excitatory and inhibitory amino acids neurotransmitter.
