阿霉素联合地塞米松抑制成纤维细胞增生的实验研究

摘要 目的研究阿霉素、地塞米松对成纤维细胞生长的抑制作用及有效浓度。为药物防治PVR提供新线索。方法应用噻唑兰比色法(MTT法)检测二者联合应用对成纤维细胞的作用。结果阿霉素明显抑制细胞生长,其50%抑制率浓度ID50=0.57mg/l;地塞米松对细胞作用呈双重性,ID50=154mg/l。联合应用,细胞增生抑制率为66.3%,比单用结果差异有非常显著性(P<0.01)。结论阿霉素、地塞米松能有效抑制成纤维细胞生长,二者合用可能有实用价值。 Objective To investigate the inhibitory activity and the effective concentraion of doxorubicin and dexamethason against the proliferation of fibroblasts of rabbit dermal in vitro,and make progress in pharmacological prevention of proliferative vitreoretinopathy. Methods Doxorubicn and dexamethason were investigated by MTT assay for their ability to their ability to inhibte rabbit fibroblast proliferation. Results Doxorubicin signifcantly suppressed the proliferation of fibroblasts in vitro. the concentraion of doxorubicin that cause 50% inhibtion (ID50) of cell growth was 0.57mg/l. Dexamthason demonstrated a bimodel effect on fibroblast . It cause inhibition of proliferation with ID50 of 154mg/l. The inhibiting effect of combination of the two drugs wan also evaluated, and it was found to be much better than in single use (IR:from 66.3% to 50%, P < 0.01).Conclusion Doxorubicion and dexamethason can effectively inhibit the proliferation of rabbit dermal fibroblast in vitro. Combination of the two drugs was thought to be of significant value possibly on treatment of PVR.
