摘要 研究了LBP2(lyciumbarbarumpolysaccharide-2)对SD大鼠腹腔巨噬细胞(MΦ)功能的影响,以探讨LBP2的免疫调节机制.MΦ吞噬中性红试验研究表明,LBP2在体外具有浓度(032~20mg/L)依赖性增强大鼠腹腔MΦ吞噬中性红的能力.另外,采用生物活性检测法测定了细胞因子的水平,将大鼠MΦ用LPS(10mg/L)和LBP2共同孵育18~24h后,培养上清及细胞内IL-1和TNF水平均随LBP2浓度的增加而增加,表明LBP2对MΦ合成和释放IL-1,TNF具有明显促进作用.LBP2对MΦ功能的影响可能是其免疫调节作用的机制之一. This research explored the effect of LBP 2 (Lycium Barbarum Polysaccharide 2) on functions of SD rats peritoneal macrophages (MΦ) and the possible immunomodulatory mechanism of LBP 2. Neutral red phagocytized by MΦ represented phagocytic activity. LBP 2 enhanced the activity of rats peritoneal macrophages to phagocytize neutral red in vitro with dose increased (0.32~20 mg/L). Moreover, the standard of cytokines was examined by the methods of biological activity. LBP 2 was shown to significantly induced intracellular and extracellular production of interleukin-1 (IL-1) and tumornecrosis factor (TNF) with LPS (10 mg/L) in 18~24 hours. LBP 2 increased the synthesis and release of IL-1, TNF from rats peritoneal MΦ in vitro. Therefore, the effect of LBP 2 on the production of IL-1, TNF from MΦ might be the main immunomodulatory mechanism of LBP 2 .
