胃癌组织P53、PCNA蛋白表达及细胞凋亡的研究

Study of expression of P53、PCNA protein and cell apoptosis in gastric carcinoma

目的探讨P53、PCNA蛋白表达和胃癌细胞凋亡关系及其它们在胃癌发生发展中的可能作用。方法采用TUNEL及流式细胞术分别检测60份胃癌、癌旁正常黏膜组织细胞凋亡和P53、PCNA蛋白表达。结果胃癌组织细胞凋亡率明显低于癌旁正常组织(<0.01);P53、PCNA蛋白在胃癌组织中的表达率明显高于癌旁正常组织(<0.01)。胃癌P53阳性组细胞凋亡率低于P53阴性组,而PCNA平均荧光强度高于P53阴性组。低分化胃癌病例其P53、PCNA蛋白高于高中分化的病例。有淋巴结转移的胃癌病例其P53、PCNA蛋白高于无淋巴结转移的病例。结论胃癌组织中癌细胞凋亡明显受到抑制,胃癌组织中P53高表达及细胞增殖活跃状态在胃癌的发生发展过程中起重要作用。

Objective To investigate the expression of P53, PCNA protein and cell apoptosis in gastric carcinoma.Methods Flow cytometry and TUNEL were used to assay cell apoptosis and expression of P53, PCNA protein in 60 cases of gastric carcinoma.Results The ratio of apoptosis was significantly lower than that in paracancer tissue（ 0.01）, and the expression of P53, PCNA protein were much higher than those in paracancer tissue.The cell apoptosis was lower and fluorescent intensity was higher in the group of positive expression of P53.The expression of P53 and PCNA in low-differentiated gastric cancer were higher than those in well-differentiated type.The expression of P53 and PCNA in gastric cancer without lymph node metastasis was lower than those with lymph node metastasis.Conclusion Apoptosis is significantly inhibited in gastric carcinoma, and increased expression of P53 protein and proliferation may play an important role in the progression of gastric carcinoma.
