地塞米松对豚鼠哮喘模型Clara细胞的影响

Effect of dexamethasone on the number of Clara cells in lung tissue in a guinea pig model of asthma

摘要 目的观察糖皮质激素对豚鼠支气管哮喘(简称哮喘)模型肺组织Clara细胞的影响。方法以29只健康豚鼠为实验动物,随机分为三组:正常对照组、过敏性哮喘组和地塞米松(DXM)预处理组。用卵白蛋白致敏、雾化建立豚鼠哮喘模型,用免疫组化的方法观察肺组织中Clara细胞,即Clara细胞蛋白10(CC10)阳性细胞及光镜下观察肺组织的病理学变化。结果过敏性哮喘组豚鼠,肺组织免疫组化CC10的表达较正常对照组和DXM预处理组减少,而且与正常对照组和DXM预处理比较,均有显著性差异(P<0.05)。讨论哮喘时肺组织中Clara细胞的数目减少,提示Clara细胞及CC10参与了哮喘的发病,它是其中一种重要的内源性的抗炎因子,糖皮质激素可增加CC10的表达,其抗炎作用可能部分是通过上调CC10在肺组织中的含量而实现的。 Objective To investigate the effect of peritoneal injected glucocorticoids on the number of Clara cells in lung tissue of a guinea pig model of asthma. Methods 29 healthy guinea pigs are randomly divided into three groups: A normal control group, an allergic asthma group and a dexamethasone (DXM) pretreatment group, in which a guinea pig asthma model was established by sensitization and challenge with ovalumin. The number of Clara cells( CC10-positive cells) in lung tissue was investigated by immunohisto- chemical method and the pathological change of lung tissue was observed in light microscope. Results The number of Clara cells in lung tissue of the allergic asthma group were significantly decreased than that of control and DXM pretreatment groups ( P 〈 0.05 ). Conclusion The decrease of Clara cell number in lung tissue of the allergic asthma group suggests that the Clara cell and CC10 may participate in the pathogenesis of asthma and work as an important endogenous anti-inflammatory factor. Glucocorticoids increased the expression of CC10 in lung tissue, and it may have an anti-inflammatory effect in the pathogenesis of asthma by increasing the CC10 levels of lung tissue.
