山药多糖对体外培养大鼠胰岛细胞活性及胰岛素分泌的影响 被引量:21

摘要 为评价山药多糖对胰岛细胞功能的影响,以不同质量浓度葡萄糖刺激胰岛素释放,通过四甲基偶氮唑盐(MTT)法检测胰岛β细胞活性,同时以反转录-聚合酶链式反应(RT-PCR)检测胰岛细胞的抗凋亡基因bcl-2的表达,研究山药多糖与大鼠胰岛细胞共培养对胰岛细胞活性与胰岛素分泌水平的影响,并对其影响机制进行探讨.结果发现,1600 mg/L组胰岛细胞活性稍有下降,但是与对照组没有显著差异(p>0.05);其余各组随培养液中山药多糖质量浓度的增加,胰岛细胞活性增加.低糖或高糖质量浓度环境中实验组与对照组的胰岛素分泌没有显著差异(p>0.05),RT-PCR发现实验组的bcl-2表达显著高于对照组(p<0.05).800 mg/L山药多糖明显提高胰岛细胞的存活率,改善胰岛功能. To explore the mechanism and the influence of chromium chloride on the viability and insulin secretion of rat pancreatic islet cell,the viability of islet β cell was measured with MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide),the insulin secretion was tested for evaluated the function of islet cell.The expression of antiapoptosis gene bcl-2 in islet cell was measured with RT-PCR(reverse transcription polymerase chain reaction).The result showed that the viability of islet cell was improved with the concentration of chromium chloride,but the ratio of apoptosis was decreased,however,the viability in the group of 1 600 mg/L chromium chloride was some decreased,and there was not significantly different(p0.05).The expression of bcl-2 was significantly higher than that of the control group(p0.05).800 mg/L chromium chloride improved the viability of islet cell.
