摘要 目的建立可用于研究工作的血清学检测方法。方法按GenBank的Fas和FasL序列设计引物,以相应质粒作阳性材料,建立各自的RT-PCR。结果27例慢性乙型肝炎血清检出sFasL-mRNA13例(48.2%),sFas-mRNA21例(77.8%)。结论目前尚无检测血清抗原的试剂、相关研究工作中可应用这一技术。 Objective To establish serological methods for detecting these soluble molecules sFas and sFasL. Methods The primerswere designed according to the published sequences of Fas and FasL. The well-defined Fas and FasL plasmids were used as positivematerials. The reverse transcription polymerase chain reaction (RT-PCR) assays were established for detecting the soluble FasL- andbas-mRNA in serum. Results The sera of 27 patients with chronic hepatitis B were tested. Thirteen (48.2%) were sFasL-mRNA,seropositive and 21 (77.8%) sEas-mRNA positive. The sFasL-mRNA might derive from liver cells or lymphocytes, and the sFasmRNA mainly from hepatocytes of hepatitis patients. Conclusions The methodology described in this paper might be used inresearch work. Combined with the in situ investigations of their membranous molecules, the functions of Fas/FasL in induction ofapoptosis may be studied more completely.
