摘要
目的检测紫杉醇药物降解材料[PLLA(左旋丙素酯)与PGA(聚乙交酯)聚合物二者的比例为9∶1]促进人脐动脉平滑肌细胞凋亡。方法将人脐动脉平滑肌细胞37℃、5%CO2条件下培养。其中一瓶,加入已消毒的紫杉醇药物降解材料(4μg/mm2),另一对照组不加入任何物质,提取细胞DNA。2%琼脂糖中电泳,紫外灯下观察结果并照相。结果本实验中紫杉醇降解材料作用于细胞一定时间后,其DNA经2%琼脂糖凝胶电泳,可见到明显的细胞凋亡"梯形带"。结论药物降解材料可导致细胞凋亡。
Objective To detect Paclitaxel(PLLA/PGA) degradable material which had been antisepsis promoting apoptosis of human's umbilical artery smooth muscle cell.Methods human's umbilical artery smooth muscle cell were cultivated in the condition of 37℃、5%CO2.Paclitaxel(PLLA/PGA) degradable material which were sterilized was added in one of bottles.Another control group was added nothing.Later the cells were refined.The electrophoresis was performed in 2% gelose.The results were observed through ultraviolet radiation light,then taken photos.Results The "echelons" of obvious apoptosis were observed in 2% gelose gel electrophoresis,after Paclitaxel(PLLA/PGA) degradable material affected cells for a while in the experiment.Conclusion Drug degradation material can promote apoptosis.
