摘要
目的:通过研究白细胞介素6(IL-6)对2型糖尿病(T2DM)大鼠骨骼肌葡萄糖转运体4(GLUT4)和糖原含量的影响,探讨其影响T2DM糖代谢的机制。方法:健康雄性SD大鼠随机分为4组:正常对照组(N组)10只,糖尿病IL-6抗原干预组(A组)10只,糖尿病IL-6抗体干预组(B组)8只,糖尿病对照组(C组)8只。测定血糖、血清胰岛素、血脂等指标;取股四头肌比色法测定肌糖原含量,免疫组化法测定骨骼肌GLUT4蛋白表达。结果:T2DM大鼠骨骼肌糖原含量和GLUT4蛋白表达较正常大鼠明显降低,IL-6抗体干预可以升高T2DM大鼠骨骼肌糖原含量和GLUT4蛋白表达。结论:T2DM大鼠骨骼肌GLUT4蛋白表达减少;IL-6抗体阻滞通过增加GLUT4蛋白表达促进骨骼肌对糖的利用。
Objective :This study is to observe the change of muscle glycogen and glucose transporter 4(GLUT4) protein content in rat skeletal muscle ceils of type 2 diabetes mellitus (T2DM) ,in order to explore the possible mechanism of interleukin-6(IL-6) influence glucose metabolism. Methods:Thirty-six healthy SD male rats Were randomly divided into four groups:10 rats with routine diets(group N), 10 rats of T2DM with introeeliae injection of IL-6 antigen diluent(group A),8 rats of T2DM with introeeliac injection IL-6 antibody diluent(group B), 8 rats of T2DM with introceliac injection of PBS(group C). The blood samples were collected to determine blood glucose ,serum insulin and lipid. The muscle glycogen was tested with colorimentry. The expression of GLUT4 in the rat skeletal muscle was tested with immunohistoehemistry analysis. Results:The content of muscle glycogen and expression of GLUT4 protein in the skeletal muscle of T2DM rat was decreased compared with that of healthy rat. IL-6 antibody block increased the content of muscle glycogen and expression of GLUT4 protein in skeletal muscle of T2DM rats. Conclusions :The expression of GLUT4 protein in skeletal muscle of T2DM hatS is lower than that in healthy rats. IL-6 antibody block increases the content of muscle glycogen by increasing the expression of GLUT4 protein in the skeletal muscle of T2DM rats.
