川芎嗪对缺氧缺血大鼠脑组织Bc1-2及Bax蛋白表达的影响 被引量:6

Effects of Ligustrazine on expressions of Bcl-2 and Bax in brain tissue of rats with hypoxic ischemic encephalopathy

摘要 目的通过观察川芎嗪注射液对缺血缺氧性脑病脑组织神经细胞形态及凋亡相关基因Bc1-2、Bax蛋白表达的影响,探讨其脑保护作用及其机制。方法将50只幼鼠分为3组,假手术组(A组)10只,模型组(B组)20只,川芎嗪注射液组(C组)20只,均于造模前腹腔注射液相应药物。应用右侧颈总动脉结扎并6%氧、94%氮混合气体处理4 h的方法制备缺氧缺血模型。3组大鼠均于脑缺氧缺血后24 h取脑组织标本检测Bc1-2、Bax蛋白表达及神经细胞凋亡情况。结果①HE染色结果显示A组海马区神经元分布均匀,形态无异常;B组海马区神经元排列紊乱,神经元肿胀变性;C组神经元镜下改变较B组明显减轻。②尼氏染色结果显示A组的海马区细胞层次清晰;B组海马区细胞数减少,神经元排列疏松;C组较B组神经元丢失明显减轻。③A组未见Bc l-2和Bax阳性细胞数增多;B组海马区Bc1-2及Bax蛋白阳性细胞数目均增多(P<0.01);与B组比较,C组海马区Bc1-2蛋白阳性细胞数目增多,Bax蛋白阳性细胞数目降低(P<0.01)。结论川芎嗪注射液可通过上调缺氧缺血大鼠脑组织中的Bc1-2蛋白表达,下调Bax蛋白表达,从而抑制神经细胞凋亡,减轻缺氧缺血所致的脑组织损伤。 Objective It is to observe the influence of Ligustrazine injection on hypoxic ischemic encephalopathy brain nerve cell morphology and protein expression of apoptosis-related genes Bc1-2,Bax,to explore its effects and mechanism of brain protection.Methods 50 young rats were divided into 3 groups,sham-operated group(group A,n=10),model group(group B,n=20) and Ligustrazine injection group(group C,n=20).The hypoxia-ischemia model was made of the right common carotid artery ligation and 6% oxygen,94% nitrogen gas mixture for 4 hours.The brain tissues of rats were taken at 24 hours after cerebral hypoxia,then the protein expressions of Bc1-2,Bax and neuronal apoptosis were observed.Results ①HE staining showed that the distribution of hippocampal neurons distributed evenly and no abnormal morphology in group A,the distribution of hippocampal neurons was irregularly and neuron was swelled and degenerated and the changes of neurons in microscopic were significantly reduced compared with the group B and in group C.②Nissl staining showed that the hippocampus at the cellular level were clear in group A,the number of cells in hippocampus were reduce and neurons loosely arranged in group B and the loss of neurons was significantly reduced in group C compared with the group B.③In group A,the number of Bcl-2 and Bax positive cells in hippocampus did not increased.In group B,the number of Bc1-2 and Bax positive cells were increased(P〈0.01).Compared with the group B,the number of Bc1-2 protein positive cells was increased,and the number of Bax protein positive cells was reduced in group C(P〈0.01).Conclusion Ligustrazine can increase the protein expression of Bc1-2 and down the protein expression of Bax in brain tissue of hypoxic ischemic rat,thus inhibit neuronal apoptosis,reduce brain damage caused by hypoxic ischemic.
