两个时间段的急性酒精性肝损伤与肝细胞凋亡 被引量:1

摘要 目的探讨两个时间段的急性酒精性肝损伤与肝细胞凋亡状况。方法将40只大鼠随机分成对照组10只和实验组30只。实验组每只大鼠按体重15 g/kg 47.5%酒精一次性灌胃。对照组每只大鼠按体重15g/kg生理盐水一次性灌胃。灌胃后分8 h和48 h分批随机取肝组织用光镜、TUNEL、电镜检测。结果实验组8 h时间段大鼠肝组织水变性、细胞凋亡病变率均为100%,无明显脂肪变性,无片状坏死;48 h时间段肝组织水变性、脂肪变性、片状坏死、细胞凋亡病变率均为100%。实验组8 h时间段凋亡肝细胞核数占肝细胞核总数比是(13.9±5.81)%,48 h时间段是(43.2±6.34)%,两组比较差异有显著性(P<0.01)。对照组大鼠未见异常病变。结论急性酒精性肝损伤48 h时间段较8 h时间段明显加重。 Objective To explore the acute alcoholic liver injury and hepatocyte apoptosis at 2 different time points. Methods Forty rats were randomly divided into experimental group ( n = 30) and control group ( n = 10 ). The experimental group was given alcohol (47.5% V/V, 15 g/kg) by garage, while NS was given in control group for replacement. Groups of rats were sacrificed for liver tissues at 8 and 48 hour. Light microscope, TUNEL and electronic micro- scope were used for pathological examination. Results Hydropic degeneration, fatty degeneration, patchy necrosis and cell apoptosis in hepatic tissues were revealed in all rats in experimental group at 48 h. Hydropic degeneration and cell apoptosis in hepatic tissues were revealed in all rats in the experimental group at 8h, while no fatty degeneration or patchy necrosis was found. The apoptosis rates of hepatocyte were 13.9 and 43.2% in the experimental group at 8 h and 48h, respectively, according to nuclear morphology. Significantly different apoptosis rates were revealed in experimental group at 8 and 48 h. No pathological change was found in control group. Conclusion Acute alcohol - induced liver injury is more severe at 48 h than 8 h in rats.
