槲皮素对人肾小管上皮细胞表达协同刺激分子B7RP-1影响的实验研究 被引量:3

摘要 目的本研究旨在观察槲皮素(quercetin)对人肾小管上皮细胞株HK-2表达协同刺激分子B7RP-1的影响,初步了解B7RP-1在肾小管上皮表达的意义及槲皮素对肾脏具有保护作用的机制。方法细胞免疫荧光(IF)方法和逆转录聚合酶链反应(RT-PCR)。结果正常的HK-2表达少量或不表达B7RP-1,在TNF-α的刺激下可以表达B7RP-1;在此基础上,在给予不同浓度的槲皮素后B7RP-1的mRNA和蛋白表达均上调,与对照组比较差异有统计学意义(P<0.05)。结论 B7RP-1在肾小管上皮细胞的表达可能对肾小管间质起保护作用;槲皮素对肾功能的保护作用可能是通过影响肾小管上皮细胞表达B7RP-1进而影响细胞免疫来实现的。 Objective To investigate the effects of quercetin on B7 related protein-1(B7RP-1)expression of human tubular epithelial cells(TECs)cultured in vitro.Through this study we aquire the elementary effects that quercetin exert on cell mediated immunity in kidney and the singnifances that B7 related protein-1 was expressed on human tubular epithelial cells.Expression of B7 related protein-1 was analyzed by reverse transcription polymerase chain reaction(RT-PCR)and cell immunnoforescence(IF).Weak or none expression of B7RP-1 was found on HK-2 cell in normal cultue.B7RP-1 expression was singficantly increased after stimulation with TNF-α;based on TNF-α,expression of B7RP-1 on HK-2 was upregulated when HK-2 was stimulated with different dose quercetin at different time points on gene and protein level.Quercetin possibly could up-regulate the expression of B7RP-1 on protein and gene level.Quercetin maybe influence the cell mediated immunity to realize the protection effect on kidney.The signifance that B7RP-1 was expressed on HK-2 maybe was to protect the tubular-interstitial.
