摘要
研究小分子药物二氯乙酸钠(dichloroacetate,DCA)联合ONYX-015对人结肠癌细胞株的体外杀伤效果,实验采用MTT法检测DCA、ONYX-015以及二者联合对结肠癌细胞株HCT116、SW620、HT29以及人正常细胞株L02增殖的抑制作用;利用Hoechst 33342染色对联合处理的细胞进行凋亡形态学观察;通过结晶紫实验检测进一步证明联合用药对肿瘤细胞的杀伤情况。实验结果表明:DCA联合ONYX-015处理4d后对结肠癌细胞株HCT116、SW620的增殖抑制率达到75%,对HT29也达到近54%的杀伤效率,并且联合治疗并未增加对人正常细胞株L02的毒性。同时结晶紫实验也证明联合治疗对肿瘤细胞的杀伤力要强于用DCA或ONYX-015单独处理。
The experiment is aimed at studying the anti-tumor effect in colorectal tumor cells by combining ONYX-015 with a small molecule dichloroacetate(DCA).First,MTT assay is used to test the growth inhibition effects of single or combination therapy on tumor cell lines HCT116,SW620,HT29 and human normal cell line L02.Second,Hoechst 33342 is added after administration in tumor cells HCT116,and 48 hours later cells are observed under fluorescent microscope.Cytopathic effect assay then is conducted to further demonstrate the efficacy of the combination therapy.Results showed that the anti-tumor efficacy of ONYX-015 is significantly enhanced in combination with dichloroacetate,while showes no overlapping toxicity against normal cell line L02.Crystal violet staining also demonstrates that the combination therapy showes better efficacy than single therapy with DCA or ONYX-015,respectively.
