摘要
膜受体是一类膜蛋白,而全长的膜蛋白很难通过基因工程获得,这给制备该类蛋白的抗体增加了难度。为了探索一条制备膜受体多克隆抗体的新途径,通过基因重组技术,将膜受体的抗原决定簇与GST标签进行融合表达,然后免疫新西兰兔,获得了相应的多克隆抗体。结果表明,利用融合GST的多肽进行抗体制备的方法是可行的。
Membrane receptors belong to a kind of membrane proteins,the full length protein of which is hard to be expressed in vitro.Therefore,it can be more difficult to obtain antibody against this kind of proteins. In order to pave a new way for preparing polyclonal antibody against membrane receptors,the antigenic determinant of membrane receptors is expressed in fusion with GST by using genetic recombination technique.The pure GST fusion polypeptide is then used to immunize a New Zealand rabbit.As a result,the polyclonal antibody is obtained.The results of ELISA and Western blot indicated that it is feasible to prepare antibody with GST fusion polypeptide.
