供体脾细胞胸腺内注射对致敏大鼠移植心脏存活的影响 被引量:1

摘要 目的:研究胸腺内耐受诱导对心脏移植(HT)存活的影响。方法:SD大鼠作为供体,Wistar大鼠作为受体。A组单做心脏移植,B组于HT前作皮肤移植致敏,C组在皮肤移植及HT前作供体脾细胞胸腺内免疫耐受诱导。结果:致敏模型能使移植排斥终点时间显著缩短,C组供心存活时间显著延长并伴相应的细胞免疫排斥指标降低。结论:未经紫外线处理的供体脾细胞也能诱导出一定程度的免疫耐受。MHC2不影响耐受诱导。该方法对细胞免疫排斥有一定的抑制作用。 Objective:To investigate the effects of intrathymic inoculation of allogeneic splenocytes on the survival of heterotopic heart transplantation(TH).Methods:SD and male Wistar rats served as skin and heart donor and recipients,respectively.Rats in group A receive HT only,rats sensitized with donor skin in group B were then challenged 7~9 days later with cardiac allografts,in group C were given intrathymic injection of donor splenocyte 7 days,before skin allografts and then HT.Results:An intrathymic injection of allogeneic spleen cells(2×10 7)prevented accelerated(<32 h) rejection in sensitized Wistar rats (group B),and prolonged the survival of SD cardiac allografts to about 8 days,this effects is donor specific and thymus dependent,it does not requires ajunctive immunosuppresive therapy and is companied by the mitigation of cell mediated immunity.Conclusion:Intrathymic injection of ultraviolet untreated allogeneic splenocytes,without the aids of immunosupressive agents could also establish a relative state of immunologic unresponsiveness to rats cardiac allografts.MHC 2 antigen does not influence the inducement of donor specific tolerance.This method can obviously depress the cell mediated immunity,but it seems that it couldn't suppress the humoral immunity and couldn't prevent the vessel injury caused by acute vascular rejection.
