摘要
目的:探讨抗CD3单抗(CD3McAb)和rIL2共同激活诱生的TAK(Tactivatedkiler)细胞的细胞毒活性本质。方法:采用MTT法分别检测TAK细胞杀伤白血病细胞的活性及其构成。结果:TAK细胞杀伤白血病细胞的活性主要由4部分组成:CD3McAb激活的CD3AK活性约占50%,rIL2诱生的LAK细胞活性约为30%,NK活性约占10%,可溶性抑制性因子的抑制活性约为10%;细胞表型分析TAK细胞主要表达活化T淋巴细胞和NK细胞的表面标志。结论:TAK细胞为异质性细胞群体。
Objective:To study the essence of the cytotoxicity of T activated killer (T AK) cells costimulated by anti CD3 monoclonal antibody (CD3McAb) and recombinant interleukin 2 (rIL 2).Methods:T AK activity against leukemia cells (K 562 cells and Raji cells) were determined separately in a MTT assay.Results:The cytotoxicity of T AK cells were composed of mainly four parts:CD3 McAb activated CD3AK activity(50%),rIL 2 induced LAK activity(30%),NK activity(10%) and the activities of inhibitory factors in T AK supernatant(10%).The analyses of phenotypes showed that T AK cells mainly expressed the surface makers of activated T lymphocytes and NK like cells.Conclusion:T AK cells are heterogeneous cell population,consisting of mainly activated T lymphocytes and NK like cells,the main part of T AK cytotoxicyty is CD3 AK activity and LAK activity.
