摘要
用高碘酸钠对普鲁兰进行氧化,制备一系列双醛普鲁兰,与胶原混合凝胶,冷冻干燥制备胶原/普鲁兰多孔复合支架材料。双醛普鲁兰的醛基和胶原的氨基反应形成希夫碱化学交联,交联度随着双醛含量的增加而提高,其降解速率低于纯胶原支架材料,且随双醛含量的增加而减小。细胞毒性毒性试验显示复合支架材料具有良好的生物相容性。表明这种复合支架材料是一种有前景的组织工程支架材料。
Collagen scaffolds have been promising to provide suitable environment for 3D cell culture in tissues engineering.However,its rapid degradation and week mechanical properties remained problems that limited many of its applications.In this study,collagen/dialdehyde pullulan complex scaffolds were prepared by mixing collagen with dialdehyde pullulan with different aldehyde contents to chemically cross-link the collagen through the formation of Schiff's base between the amino on collagen and the aldehyde on pullulan.With higher aldehyde content in pullulan,the complex scaffold thus formed showed improved degradation resistance.The MTT assay indicates that the cell proliferation on the complex scaffolds had not different with that on the pure collagen scaffold.These complex scaffolds are suitable for 3D cell culture in tissue engineering.
