摘要
目的:观察活化B淋巴细胞与NS1骨髓瘤细胞融合瘤苗对荷NS1瘤BABL/C小鼠免疫治疗作用。方法:用灭活NS1瘤细胞及其裂解物免疫BALB/C小鼠活化的B淋巴细胞;以50%PEG为融合剂,将免疫脾B淋巴细胞与野生型NS1骨髓瘤细胞融合,经HAT选择性培养、筛选融合细胞,以融合细胞作为瘤苗,免疫治疗荷NS1瘤小鼠2次。结果:融合细胞体在宿主体内的致瘤性降低;融合细胞瘤苗免疫治疗荷瘤小鼠的肿瘤组织体积逐渐减小、最终完全消失,小鼠存活期显著延长。结论:活化的B淋巴细胞与NS1细胞融合瘤苗能有效诱导小鼠体内的抗肿瘤反应。
Aim: To prepare a tumor vaccine by fusion of activated B cells with NS 1myeloma cells and investigate its antitumor immunotherapeutic effects in the NS 1myeloma bearing BALB/C mice. Methods:The activated B cells from the spleen of BALB/C mice immunized with deactivated NS 1myeloma cells and lysates were fused with wild live NS 1myeloma cells by 50% PEG protocol.The resulting fusion cells were selected by HAT medium. The mice loaded NS 1 myeloma cells under their skin were treated with the fused cells twice weekly.Results: The tumorigenicity of the fused cells became decreased apparently,in contrast to that of NS 1cells. The model tumor volumes gradually became small or disappeared at last. The survial time of tumor bearing mice markedly prolonged. Conclusion: The findings of this study indicate the vaccine based on fusion of the acitivated B cells with NS 1 myeloma cells can induce effectively in vivo antitumor immune responses of mice.
