摘要
研究rasp21和p53表达和DNA倍体异常与尿路上皮肿瘤发生关系。方法:应用流式细胞术和细胞免疫荧光技术对52例尿路上皮肿瘤rasp21和p53蛋白和DNA含量进行定量检测。结果:异倍体22例(42.3%),二倍体30例(57.7%);异倍体肿瘤其DNA指数、增殖指数和S期细胞比率均高于二倍体肿瘤,但差异无显著性(P>0.05);rasp21和p53共同表达25例(48.1%),随肿瘤病理分级分期的上升而增加(P<0.05);异倍体肿瘤rasp21和p53共同表达16例(72.7%),二倍体肿瘤rasp21和p53共同阳性9例(30%),两组间差异显著(P<0.01)。结论:DNA倍体异常、rasp21和p53的共同表达参与尿路上皮肿瘤的发生和发展。||关键词##4尿路上皮肿瘤;;rasp21;;p53;;DNA倍体;;
Objective: To study the relationship between the abnormal ras p21, p53 expression,DNA ploidy and the carcinogenesis of urothelial tumors. Methods: The ras p21, p53 protein expressionand DNA content were quantitatively deternlined using flow cytometry and cellular immunofluorescence tech-nique in paraffin embedded tissues from 52 cases of urothelial tumors. Results: 42. 3% (22/52) of aneu-ploidy and 57. 7% (30/52) of diploidy were observed in 52 cases of urothelial tumors. The DNA index,proliferation index and S - phase fraction in aneuploidy tumors were higher than those in the diploidy tu-more, but the difference was not significant ( P > 0. 05) . Co - positive expression of ras p21 and P53were seen in 25 cases (48. 1% ) and it increased along with the escalation of the pathological grading(P<0.05) . Coexpression of ras p21 and p53 observed in aneuploidy tumors was 72.7% ( 16/22)and in diploidy was 30% (9/30) with significant difference ( P<0.01) . Conclusion: DNA ploidyabnormality and co - expression of ras p21 and p53 play an important role in the development of urothe-lial tumors.
