摘要 目的使用分子排阻色谱(SEC)法测定玻璃酸钠(SH)相对分子质量(Mr)及其分布。方法考察进样浓度、柱温及流速对SEC法测定SH的Mr及其分布的影响。结果进样浓度对高分子SH的色谱峰形及测定结果影响较大,降低进样浓度色谱峰形明显改善,且测定结果精密度良好;柱温对SH的Mr测定结果有一定的影响;流速对SH的Mr及其分布基本无影响。结论在SH进样浓度0.1 mg/mL,柱温30℃,流速0.5 mL/min的色谱条件下,用SEC法测定SH的Mr及其分布的色谱峰形良好,结果准确可靠,可用于SH的质量控制和应用研究。 Objective To determine the relative molecular mass (Mr) and its distribution of sodium hyaluronate (SH) by the size exclusion chromatography (SEC). Methods The influences of sample concentration, column temperature and flow rate on the determination of Mr and its distribution by SEC were studied. Results For SH samples with high molecular weight, the concentration had significant influence on the determination of Mr and its distribution by SEC. Good peak shapes and determination results could be obtained by decreasing the concentration of SH. Column temperature had some influence on the determination of Mr and its distribution by SEC, but flow rate had no influence. Conclusion The sample concentration of 0.1 mg/mL, column temperature of 30 ℃ and flow rate of 0.5 mL/min are suitable for the determination of Mr and its distribution of SH by SEC, which can be used for the quality control and application research on SH.
