摘要
目的为探索胆碱能神经元的胚胎来源,验证放射状胶质细胞是否表达乙酰胆碱转移酶。方法通过对孕14天(E14)大鼠胚胎端脑冠状石蜡切片进行波形蛋白(放射状胶质细胞标志性蛋白,vimentin)和乙酰胆碱转移酶(胆碱能神经元的标志性蛋白ChAT)双重荧光免疫组织化学染色,并通过重叠同一视野内的荧光双染的图片来精确地体现放射状胶质细胞表达ChAT的位置。结果 E14大鼠端脑腹侧脑室区有部分组织呈vimentin/ChAT免疫化学双染阳性,这和资料中证实的胆碱能神经元E14天出现于第三脑室的腹背侧部的结论是一致的。结论 E14大鼠端脑内的放射状胶质细胞表达ChAT。表明放射状胶质细胞具有神经干细胞性质,它不断地进行不对称分裂,产生胆碱能表型的成神经细胞,即胆碱能前体细胞。
Objective To explore the embryonic source of cholinergic neurons and to verify whether the radial glial cells can express acetylcholine transferase.Methods Coronary paraffin sections of rat telencephalon on embryonic 14 days(E14) were prepared.Double fluorescent and immunohistochemical staining of vimentin(a protein marker of radial glial cell) and choline acetyltransferase(ChAT,a protein marker of cholinergic neuron) to test whether ChAT was expressed in the population of radial glial cell in vivo.Overlapped pictures in the same field of vision reflected the precise position where the radial glial cells expressed ChAT.Results Vimentin-positive cells coexpressed ChAT in ventral ganglionic eminence in E14 rats,which was consistent with the conclusion in previous reports that cholinergic neurons emerged in the dorsal or ventral side of the third ventricle on the E14 day.Conclusion In the E14 rat telencephal brain ChAT was expressed in the radial glial cell population.This result suggested that radial glial cells had the property of neural stem cells and continually underwent asymmetrical cell division to generate neuronblasts with cholinergic phenotype,or cholinergic precursors.
