血府逐瘀汤对兔椎间盘退变模型软骨组织中TNF-α的影响

Experimental study on the effect of Xuefuzhuyutang on TNF-α in the cartilage tissue of disc degeneration of rabbits

目的:观察肿瘤坏死因子(TNF-α)在椎间盘退变中的作用和血府逐瘀汤对其的影响。方法:①取10只6月龄日本长耳白兔,随机分手术造模组与假手术组,每组5只,术后8周证实造模成功后分别处死取软骨终板,用Elisa法测TNF-α含量变化。②取20只6个月龄日本长耳白兔按上述手术造模组方法造模,造模成功后随机分为生理盐水组和血府逐瘀汤组,每组10只,予相应药物治疗。治疗后第1、2个月分别用Elisa法测软骨终板TNF-α含量变化。结果:①手术造模组较假手术组椎间盘软骨中TNF-α明显升高,差异有统计学意义(P<0.05)。②治疗1个月后血府逐瘀汤组与生理盐水组比较,椎间盘软骨中TNF-α降低明显,差异有统计学意义(P<0.05);治疗2个月后血府逐瘀汤组较生理盐水组比较,椎间盘软骨中TNF-α明显降低,差异有统计学意义(P<0.05);手术造模组与生理盐水组比较,差异无统计学意义(P>0.05)。结论:血府逐瘀汤能降低退变椎间盘软骨组织中TNF-α水平。

Objective：To observe the changes of tumor necrosis factor alpha（TNF-α）in the disc degeneration and normal tissues and to study the impact of Xuefuzhuyutang on TNF-α in the cartilage tissue of disc degeneation.Methods：i.Ten 6-month-old Japanese white rabbits were randomly divided into operation model group and sham operation group,5 in each and were executed in 8 weeks after operation when successful modeling was proved.Cartilage end-plate was taken out to measure TNF-α content with the ELISA method.ii.Another 20 6-month-old Japanese white rabbits followed the above-mentioned operation of the operation model group randomly divided into normal saline group and Xuefuzhuyutang group when successful modeling was proved,10 in each.The content of TNF-α was determined in 1 and 2 months after treatment with the same method.Results：i.The content of TNF-α in the operation model group was higher than that in the sham operation group,with a statistically significant difference（P0.05）.ii.The content of TNF-α was gradually decreased in the Xuefuzhuyutang group;both the levels in 1 and 2 months after treatment were lower that those in the normal saline group（P0.05）;no significant difference was noted between the operation model group and the normal saline group（P0.05）.Conclusion：Xuefuzhuyutang may suppress or delay intervertebral disc degeneration by down-regulating the content of TNF-α in cartilage endplate tissues of the degenerated lumbar disc.
