G-CSF和GM-CSF对完全缓解AML患者树突状细胞亚群影响的体内外研究 被引量:2

摘要 目的研究细胞因子干预下树突状细胞(DC)功能及其分化亚群。方法获取完全缓解的AML患者DC细胞,流式细胞学和形态学方法进行鉴定。MTT法检测其增殖活性和促淋巴细胞增殖活性。ELISA法和PCR技术检测IL-12的水平。结果在G-CSF和GM-CSF干预后,DC细胞高表达HLA-DR(67±5)%、CD11c(58±9)%,其促淋巴细胞增殖能力活性增高,且呈时间依赖性。G-CSF干预组移植物抗宿主病(GVHD)轻于GM-CSF组。结论G-CSF和GM-CSF联合干预后DC细胞的增殖及促增殖活性增强,但G-CSF可减轻小鼠GVHD的发生,可能与G-CSF、GM-CSF可以调节DC细胞的分化亚群相关。 Objective It is to study the function of dendritic cell(DC) under the interference of cytokine and its subset splitted.Methods The DC cells of AML patients who reached completed remission were obtained and evaluated with flow cytology and morphology methods.The proliferated activity and promoting lymphocyte proliferated activity of DC cells were detected with MTT method.The level of IL-12 was detected with ELISA method and PCR technique.Results After interference of G-CSF and GM-CSF,DC cells high expressed HLA-DR(67±5)% and CD11c(58±9)%.The promoting lymphocyte proliferated activity was heightened and which was showed time dependence.The GVHD in G-CSF group was lighter than that in GM-CSF group.Conclusion The proliferated activity and promoting lymphocyte proliferated activity of DC cells can be enhanced after the interference of G-CSF and GM-CSF,but G-CSF can lighten the occurrence of GVHD,which may be concerned with that G-CSF and GM-CSF can regulate the subset splitted of DC cells.
