野生毛葡萄单芽茎段的离体快繁培养 被引量:1

摘要 以野生毛葡萄花溪3号和鼎罐城1号的单芽茎段为外植体,比较了离体培养时腋芽诱导和生根的差异,并对腋芽诱导和生根培养基进行了筛选。结果表明:适宜野生毛葡萄花溪3号和鼎罐城1号的腋芽诱导培养基为MS+6-BA 1.0 mg/L+IAA 0.1 mg/L,诱导率分别为76.34%和85.89%;适宜的继代与生根培养基为1/2 MS+6-BA 0.01 mg/L+IBA(0.1-0.2)mg/L+IAA(0.1-0.2)mg/L。培养的试管苗主根较多、较长且粗壮,苗木基部无愈伤组织,叶色浓绿,植株生长健壮。 The stem with single bud of Huaxi 3 and Dingguancheng 1,two wild Vitis quinquangularis varieties,were cultured on different induction and rooting media to study the induction rate of their axillary bud and rooting rate for screening the optimum media.The results showed that the induction rate of axillary bud of Huaxi 3 and Dingguancheng 1 stem sections cultured on MS+1.0mg/L 6-BA+0.1mg/L IAA was 76.34% and 85.89% respectively,the optimum medium for subculture and rooting of Huaxi 3 and Dingguancheng 1was 1/2MS+0.01mg/L 6-BA+0.1~0.2mg/L IBA+ 0.1~0.2mg/L IAA,the tube seedlings cultured on the optimum media for their subculture and rooting had more longer and thicker main roots,no callus on their basis and thick green leaves.
