摘要
为培育盾叶薯蓣优良品种,培养盾叶薯蓣优质种苗,促进盾叶薯蓣的规模化生产。对高皂素含量的盾叶薯蓣株系进行了组织培养与快繁研究。结果表明,茎尖外植体产生愈伤组织的时间短、出愈率高。在添加0.5 mg/L BA和0.5 mg/L NAA的培养基上,出愈率接近70%,愈伤组织状态良好;1.0 mg/LKT和0.2 mg/L NAA组合可使愈伤组织的芽分化率达90.9%;在添加0.5 mg/L NAA和1.0~2.0 mg/LBA的培养基上,腋芽增殖率最高,且生长健壮。已伸长的腋芽转移至不加任何激素的MS培养基上即可生根,形成完整植株。
The explants of a Dioscorea zingiberensis line with high saponin content were cultured and propagated to breed good cultivars and high quality seedlings for scale production of Dioscorea zingiberensis.The results showed that the callus culture days and callus rate induced from shoot tip explants was minimum and maximum respectively,the callus rate of shoot tip explants cultured on MS+ 0.5 mg/L BA+0.5 mg/L NAA reached nearly 70% and its callus status was good,The bud differentiation rate of calli cultured on MS+1.0 mg/L KT+0.2mg/L NAA was up to 90.9% and the optimum medium for propagation of axillary buds was MS+0.5mg/L NAA +1.0 or 2.0 mg/L BA.The elongated axillary buds cultured on MS medium could produce roots and regenerate intact plantlets.
