摘要 为了优化出厚朴顶芽愈伤组织诱导的最适培养条件,以MS为基本培养基,探讨了6-BA、NAA、2,4-D以及光照对厚朴顶芽诱导的影响。结果表明:厚朴顶芽诱导的最适培养基为MS+2.0 mg/L2,4-D+0.5 mg/L 6-BA+1.0 mg/L NAA+30 g/L蔗糖+8 g/L琼脂,pH 5.8,培养13 d后,顶芽诱导出愈伤组织。诱导出的顶芽愈伤组织在温度(25±2)℃,光周期为12h/d,光照强度为1 500~2 000 lx条件下进行光培养,其比暗培养[(25±2℃)]差。 The terminal buds of Magnolia officinalis were cultured on MS with different combinations of 6-BA,NAA and 2,4-D under different light condition to study the effect on its callus induction and optimize the optimum culture condition for callus induction of Magnolia officinalis terminal buds.The results showed that the optimum medium for callus induction of Magnolia officinalis terminal buds is MS+2.0 mg/L 2,4-D+0.5 mg/L 6-BA+1.0 mg/L NAA+30 g/L sucrose+8 g/L agar at pH 5.8 and the callus can be induced from Magnolia officinalis terminal buds cultured on the optimum medium after 13 d.The callus induction rate of Magnolia officinalis terminal buds cultured on the optimum medium under the condition of(25±2)℃ and darkness is higher than that under the condition of(25±2)℃,12 h/d photoperiod and 1 500~2 000 lx light intensity.
