摘要 目的利用体外培养重建同基因角膜上皮组织的新方法,为角膜化学伤后重建角膜表面提供理想的角膜上皮移植片。方法以同种或异种脱水保存的浅层角膜基质片作为载体,通过体外培养自体角膜缘上皮细胞,并使其在体外形成复层角膜上皮组织。结果经组织学、细胞超微结构的观察,以及细胞分化蛋白表达的检测,证实体外培养的角膜上皮组织与生理状态下的角膜上皮组织相近。结论体外培养的角膜上皮组织片可作为较理想的角膜上皮移植材料。 ObjectiveTo provide corneal epithelial transplant for corneal surface reconstruction after chemical or thermal injuries of eyes,autologous limbal cells were cultured in vitro for use as transplants.MethodsThe upper parts of corneal stroma preserved in glycerine was used as a carrier for growing autologous limbal corneal epithelial cells in vitro.ResultsStratified corneal epithelium was successfully cultured in vitro.Histological,ultrastructural and immunohistochemical study of the cultured stratified squamous corneal epithelium showed that the cultured corneal epithelium was quite similar in histological structure,ultrastructure and expression of differentiation related protein to those in vivo.ConclusionTherefore,the cultured corneal epithelium may prove to be potential grafts for corneal epithelium transplantation.
