高眼压鼠视网膜胶质纤维酸性蛋白免疫组织化学观察

An immunohistochemical study of glial fibrillary acidic protein in rat’sretinal Müller cells under acute ocular hypertension

目的通过观察高眼压后Ｍüｌｅｒ细胞胶质纤维酸性蛋白（ＧＦＡＰ）的表达情况，探讨Ｍüｌｅｒ细胞在高眼压性视网膜损伤中的作用及意义。方法前房加压灌注法制作大鼠急性高眼压模型，免疫组织化学方法显示视网膜上ＧＦＡＰ的表达，并采用计算机图像分析系统加以定量。结果急性高眼压后视网膜上ＧＦＡＰ阳性染色迅速增加，一段时间后渐消退。结论高眼压状态可诱导Ｍüｌｅｒ细胞发生反应性胶质化，提示Ｍüｌｅｒ细胞可能在高眼压损伤过程中发挥保护作用。

ObjectiveTo investigate the role and significance of the Müller cell in glaucomatous damage by observing expression of glial fibrillary acidic protein (GFAP) in Müller cells under acute ocular hypertension.MethodsThe animal model of acute ocular hypertension was made by perfusing physiological saline into the anterior chamber of rats.The GFAP expression in retina was shown with immunohistochemistry,and was quantitated by computerized morphometry.ResultsGFAP expression increased rapidly following acute ocular hypertension,and decreased gradually after a period of time.ConclusionAcute ocular hypertension can induce Müller cells to undergo reactive gliosis.It suggests that the Müller cell may play an important role in protecting the retina from ocular hypertension.
